The branch, currently at 220 High Street, Methil, is relocating to "better, brighter premises" at 204-206 High Street.
Royal Mail bosses said public consultation and feedback had been in favour of a move, which would see customers benefit in a nu
mber of ways.
There will be a brand-new, modern, purpose-built serving counter and an open-plan format, offering retail services alongside Post Office products and services, in addition to on-line Lottery.
There will be level entry to the branch for ease of access, which should help customers with mobility constraints.
The current branch will close at 12.30pm on July 27 and the new one will open at 9am on Wednesday, July 29.
Helen Belford, field change advisor with Post Office Ltd., said: "The facilities and service we provide to our customers is of paramount importance and we are confident our customers will benefit, in the longer term, by this change.
A Royal Mail spokesman said customers and "opinion formers" were invited to offer comments and feedback on any issues, particularly relating to the operational aspect of the new location.
They were quizzed on proposed opening hours, access and facilities at the new site, and any local issues specific to Methil that should be considered.
"We received two responses, one from a customer and one from a Member of the Scottish Parliament, both of whom were in support of the move," added the spokesman.
"The branch had been run by a temporary agent since our contract with the former sub-postmaster came to an end.
"Following our selection and appointment process, a new sub-postmaster was appointed who had offered alternative premises."
During the short closure period, customers requiring Post Office facilities can use any convenient Post Office service, with two of the closest being the Crossroads branch, at 285 Methilhaven Road, Methil, and the Buckhaven outlet at 106-110 Wellesley Road, Buckhaven.
